Telah dilakukan penelitian mengenai optimasi pembentukan kapsula pada Salmonella typhii sebagai bahan baku produksi vaksin demam Tifoid (Vi Capsular Polysaccharide). Selama ini, vaksin di Indonesia diimpor dari luar negeri dan dijual dengan harga yang relatif tinggi. Maka dari itu, dilakukanlah pembuatan vaksin demam tifoid dari Indonesia sebagai solusi terhadap permasalahan penyediaan vaksin demam tifoid di Indonesia. Optimasi dilakukan dengan memodifikasi medium dengan menambahkan substrat (glukosa) yang konsentrasinya diatur dan dijaga pada 2g/L, 3g/L, 4g/L, 5g/L, sementara pH diatur dan dijaga konsentrasinya sedemikian rupa pada variasi pH 6,7 dan 8. Semua perlakuan diberi agitasi sebesar 200 rpm pada incubator shaker dengan suhu 32°C. Selama proses fermentasi berlangsung, diamati perubahan jumlah sel Salmonella typhii (optical density), perubahan pH, serta perubahan konsentrasi glukosa. Setelah data diperoleh, dilakukan analisis berupa perhitungan laju perubahan biomassa, laju perubahan pH dan laju perubahan konsentrasi glukosa. Analisis jumlah kapsula yang dihasilkan juga dilakukan dengan metode ELISA. Berdasarkan analisis tersebut, didapatkan hasil dari proses fermentasi fed batch dengan optimasi konsentrasi glukosa yang dijaga pada 3g/L dan konsentrasi pH 6, 7, dan 8 berturut-turut berupa laju pertumbuhan sebesar 7,6 x 107 sel/jam dan konsentrasi kapsula 3.4 μg/mL, laju pertumbuhan 2,5 x 108 sel/jam dan konsentrasi kapsula 16.5 μg/mL dan laju pertumbuhan 4,1 x 108 sel/jam dan konsentrasi kapsula 35.3 μg/mL. Dari hasil berikut, dapat disimpulkan bahwa dengan mengoptimasi medium pada konsentrasi glukosa 3g/L dan pH 8 , dihasilkan biomassa sel terbanyak serta jumlah kapsula tertinggi dengan laju pertumbuhan 4,1 x 108 sel/jam dan konsentrasi kapsula 35.3 μg/mL.

Description Alternative :

Research has been conducted on Salmonella typhi capsule production as basic material for typhoid fever vaccine production (Vi Capsular Polysaccharide) . In Indonesia, typhoid fever vaccines were imported and sold at relatively high prices. Therefore, typhoid fever vaccine production were conducted in Indonesia as a solution to supplying cheap and effective vaccines in Indonesia. Modification of growth medium were done by optimizing its substrate intake and pH concentration. Substrate were given at a steady and maintained concentration at 2g/L, 3g/L, 4g/L and 5g/L. pH concentration was also maintained at pH 6 , 7 and 8. All optimization process were given agitation of 200 rpm and stored in 32°C incubator shaker . During the fermentation process, bacterial cell growth , pH changes and glucose concentration changes were observed respectively with spectrophotometer, pH meter and AccuCheck™. After the data were obtained, cell growth rate, pH changes rate and glucose concentration changes rate were analyzed. Capsule production from pH optimization fermentation were analyzed using ELISA method. Based on the analysis, the result of fed batch fermentation process with optimum glucose concentration maintained at 3g / L and pH concentration 6, 7, and 8 respectively in the form of growth rate of 7,6 x 107 cells / hour and average capsule concentration of 3.4 μg / mL capsule, Growth rate of 2,5 x 108 cells / hour and average capsule concentration of 16.5 μg / mL and growth rate 4,1 x 108 cells / hour and average capsule concentration 35.3 μg / mL. From the following results, it can be concluded that by optimizing the medium at 3g / L glucose concentration and maintaining pH at 8, the highest cell biomass was produced and the highest capsule concentration were obtained with growth rate of cell was 4,1 x 108 cells / hour and average capsule concentration of 35.3 μg / mL.
